چکیده

Thiamine diphosphate functions as a cofactor in enzymes that catalyze essential reactions in metabolic pathways of all organisms. Phosphoketolase is a thiamine diphosphate-dependent key enzyme in the catabolism of sugar in various microbes. In particular, this enzyme is indispensable for unusually high effi ciency of energy production of the glycolytic pathway of benefi cial commensal bacteria, bifi dobacteria. However, its structure and mechanism have long been enigmatic. We determined the three-dimensional structure of phosphoketolase from Bifi dobacterium breve. Details of its unique reaction mechanism involving dehydration and nucleophilic attack by phosphate were revealed based on “snapshot” X-ray crystal structures.

Crystal structures of phosphoketolase: thiamine diphosphate-dependent dehydration mechanism.

Thiamine diphosphate (ThDP)-dependent enzymes are ubiquitously present in all organisms and catalyze essential reactions in various metabolic pathways. ThDP-dependent phosphoketolase plays key roles in the central metabolism of heterofermentative bacteria and in the pentose catabolism of various microbes. In particular, bifidobacteria, representatives of beneficial commensal bacteria, have an e...

Expression of the xylulose 5-phosphate phosphoketolase gene, xpkA, from Lactobacillus pentosus MD363 is induced by sugars that are fermented via the phosphoketolase pathway and is repressed by glucose mediated by CcpA and the mannose phosphoenolpyruvate phosphotransferase system.

Purification of xylulose 5-phosphate phosphoketolase (XpkA), the central enzyme of the phosphoketolase pathway (PKP) in lactic acid bacteria, and cloning and sequence analysis of the encoding gene, xpkA, from Lactobacillus pentosus MD363 are described. xpkA encodes a 788-amino-acid protein with a calculated mass of 88,705 Da. Expression of xpkA in Escherichia coli led to an increase in XpkA act...

Metabolism of a plant derived galactose‐containing polysaccharide by Bifidobacterium breve UCC2003

In this study, we describe the functional characterization of the Bifidobacterium breve UCC2003 gal locus, which is dedicated to the utilization of galactan, a plant-derived polysaccharide. Using a combination of molecular approaches we conclude that the galA gene of B. breve UCC2003 encodes a β-1,4-endogalactanase producing galacto-oligosaccharides, which are specifically internalized by an AB...

Characterization of the D-xylulose 5-phosphate/D-fructose 6-phosphate phosphoketolase gene (xfp) from Bifidobacterium lactis.

A D-xylulose 5-phosphate/D-fructose 6-phosphate phosphoketolase (Xfp) from the probiotic Bifidobacterium lactis was purified to homogeneity. The specific activity of the purified enzyme with D-fructose 6-phosphate as a substrate is 4.28 Units per mg of enzyme. K(m) values for D-xylulose 5-phosphate and D-fructose 6-phosphate are 45 and 10 mM, respectively. The native enzyme has a molecular mass...

Thiamine hydrochloride (Vit-B1): An optimized green alternative for the synthesis of polyhydroquinoline derivatives

Thiamine hydrochloride (Vitamin-B1) has been utilized as an effective biodegradable catalyst for the one pot synthesis of polyhydroquinoline derivatives via four component reaction of dimedone, aldehyde, ethylacetoacetate and ammonium acetate in ethanol under mild conditions. The method presented here offers several advantages over reported procedures in terms of environmentally benign catalyti...
